Abstract

Chemokines are constitutively expressed by neurons and glia in the CNS. SDF‐1, ligand for CXCR4, plays a critical role in neuronal migration and axon pathfinding. On the other hand, NCAM ‐Neural Cell Adhesion Molecule shows a post‐translational modification, PSA‐NCAM, that regulates neural cell‐cell interactions. We studied whether SDF 1 alpha and Fractalkine could regulate NCAM and PSA‐NCAM protein levels in cortical neurons 6 DIV overnight treated with these recombinant chemokines. Our results displayed that fractalkine donwregulated PSA‐NCAM and NCAM 140 protein levels vs their controls. Interestingly, axonal branching and PSA‐NCAM inmunofluorescence decreased after fractalkine treatment. Besides, we evaluated whether chemokines could play a role in learning and memory. SDF1 alpha inmunoreactivity increased in the hippocampus and prefrontal cortex of rats with improved behavioural performance. Interestingly, AMD3100 ‐CXCR4 antagonist‐ infusion before learning, impairs memory consolidation. In conclusion, these findings suggest a role of fractalkine as cell adhesion molecule in the CNS throuough PSA‐NCAM regulation in the rat prefrontal cortex “in vivo” and “in vitro”
