Simplified method for microscopic determination of Giardia disinfection efficiency

Abstract
Methods used in the laboratory for the excystation of cysts of Giardia muris are described. An account of their enumeration by the standard method using phase contrast microscopy and a modified volumetric method in which the number of trophozoites was determined by microscopic examination and the volume of sample from which these trophozoites were derived was calculated is given. A statistical evaluation of the results showed there was no significant difference between the log survival figures determined by the 2 methods, based on the Wilcoxson signed rank test. Qualitatively there was also good agreement between the 2 methods over the entire range of inactivation treatments used during the study, which involved the application of 3 different disinfectants and 3 different source waters. For disinfection studies the volumetric and standard methods were judged to give results which were indistinguishable.
