Anatomic-functional correlates of adenosine-5' triphosphate triggered vagal depressor reflex

Abstract
Adenosine-5’-triphosphate (ATP) plays a crucial role in intracellular energetics and metabolism. Once outside of the cells, however, ATP exerts pronounced effects on many organs and tissues, including the cardiovascular system [1]. Extra-cellular ATP exerts negative chronotropic and dromotropic actions on sinus node automaticity and atrioventricular (AV) nodal conduction, respectively [2]. A decade ago we had established the role of the vagus nerve in mediating, in part, these actions of ATP. Specifically, we have shown that ATP is more potent than adenosine in suppressing the sinus and AV nodes, and that this difference between the two compounds is due to the ability of ATP to abruptly and transiently increase vagal input to the heart [3]. Indeed, when the vagal effect is blocked by either surgical or pharmacologic perturbations, there is no difference between the negative chronotropic and dromotropic actions of ATP and adenosine [3]. The vagal component of ATP’s action exhibits species variability, that is, it is manifested in cat, dog, and human but not in rat, guinea pig, and rabbit [2]. Thus, using canine models in vivo we have confirmed the vagal component in the cardiac electrophysiologic actions of ATP. Specifically, physostigmine and elevation of plasma Ca2+ levels enhanced the actions of ATP, while atropine and verapamil attenuated these actions [3–6]. In addition, prostaglandins, the synthesis and release of which are triggered by ATP, do not mediate the negative chronotropic and dromotropic actions of ATP [7].
