Abstract
The blood–brain barrier (BBB) serves as an interface between circulating blood and the brain interstitium. The BBB can respond to and be influenced by alterations in the internal and external conditions of the human body as well. Disruption of BBB integrity mediates the pathophysiology of a wide range of central nervous system (CNS) disorders and the neurological sequelae of many systemic diseases and disturbances. This article reviews the pathology of the BBB in various disturbances and stresses intrinsic and extrinsic to the CNS. A thorough understanding of the pathophysiology of the BBB and of the factors influencing its structure and function is essential for the development of targeted drug delivery strategies and safer and more effective therapies in the management of many diseases of the CNS.
