Abstract

We have measured the pH-dependence of the depolarization ratio and the excitation profiles of two prominent Raman lines (1375 cm −1, 1638 cm −1) of oxyhaemoglobin in the resonant region between the Q- and B-absorption bands. From these data pK-values of pH-induced conformational changes of the protein, which by haem-apoprotein interaction also change the conformation of the prosthetic porphyrin, can be extracted. In comparison to these data we have investigated the pH-dependence of the extinction coefficients in the absorption maxima of the Q- and B-bands. Their behaviour can be explained by the same mechanism.
