Planar co-culture of rabbit chondrocytes and bone marrow stromal cells: screening of their culture ratio

Background: Bone marrow stromal cells can differentiate into chondrocytes under certain conditions, but in-depth research on the co-culture of chondrocytes and bone marrow stromal cells in a planar manner is still lacking. Objective: To clarify the optimal ratio of bone marrow stromal cells to chondrocytes in co-culture, cell proliferation activity, the expression patterns of chondrocyte-specific proteins, and the optimal time and passage number for cell transplantation. Methods: Chondrocytes and bone marrow stromal cells were isolated and cultured, and after passage, they were divided into 5 groups: chondrocyte-only group; co-culture group; chondrocyte-to-bone marrow stromal cell group (7:3, 5:5, 3:7); and bone marrow stromal cell-only group. After passage to the 4th generation (G4), cell morphology of each group at different passage stages was observed using an inverted phase-contrast microscope. The TT assay was used to detect cell proliferation activity, and toluidine blue and alicin blue staining and immunocytochemistry were used to detect type II collagen expression. Results and conclusions: Cell phenotypes were normal in all co-culture groups at all time stages. Cells in generations G1-G3 showed active proliferation, while cell proliferation decreased in generation G4. Toluidine blue and alicin blue staining and type II collagen expression were higher in generations G2 and G3. Based on all indicators, a 5:5 or 3:7 ratio of chondrocytes to bone marrow stromal cells was considered optimal. Planar co-culture of chondrocytes and bone marrow stromal cells maintained the morphology and protein expression characteristics of chondrocytes. For cell transplantation, a 5:5 or 3:7 ratio of chondrocytes to bone marrow stromal cells is optimal.
