Abstract

1. 1. The observation that hepatic cholesterol synthesis exhibits circadian variation in the fed and fasted rat was confirmed. Additionally, diurnal determinations were made of hepatic cholesterol and bile acid content, fatty acid synthesis, and plasma free fatty acid levels. Rhythmic variations in all these parameters were found. Hepatic cholesterol and bile acid content rose through the early evening, the cholesterol reaching a peak at midnight, the bile acids at 9 p.m. Both cholesterol and bile acid contents were higher in the fasted animal. Plasma free fatty acid was rather stable throughout the day other than a peak at 3 and 6 p.m. in the fed animal. The values were much higher in the fasted animal. Hepatic fatty acid synthesis also exhibited a daily rhythm as previously reported. This appeared to vary reciprocally with the free fatty acid levels. 2. 2. The data are interpreted in the light of the documented variation of levels of β-hydroxy-β-methyl-glutaryl-CoA reductase, and it is suggested that the rise in plasma free fatty acid may be the signal for the increase in the amount of this enzyme, and that the bile acids may represent a represser of its further synthesis.
